Bass help?

I recently bought a brand new 4 string bass, never messed with the strings other than tuning them. When I play an open note, it sounds fine. But when I press on a fret it sounds all twangy like as if the strings are hitting the other frets. Does anyone know what's causing this or how I could fix it? Also, what settings do you use for your amps to get best sound? Thanks.
